Ad Targeting is finding the best ad to show a visitor. Ad Position is finding the best combination of content and ads on each page (and for every screen size).
The only way of knowing which layout is 'better' is to define your goals (user experience, revenue, or a balance of the two), and TEST to find out. Every site is different, continuous testing is the only way to know for sure.
