Ad targeting isn't the same as Ad location. The Ezoic system tests lots of different ad positions and sizes on a page. Each layout test gets tracked for UX (user experience metrics - time on site, page views per visitor and bounce rate) and also for the income per thousand visitors (epmv). Ad Targeting is finding the best ad to show a visitor. Ad Position is finding the best combination of content and ads on each page (and for every screen size).
The only way of knowing which layout is 'better' is to define your goals (user experience, revenue, or a balance of the two), and TEST to find out. Every site is different, continuous testing is the only way to know for sure.
